A Computational Approach to Predict the Cardiac Alternans of Action Potential Duration on Electrophysiological Responses

Alternans is the periodic beat-to-beat alternations in the shape of cardiac electrical signals, such as action potential duration (APD; APD alternans). A preliminary study explains that decreasing only the ryanodine receptor (RyR) inactivation rate constant (kiCa) induced the generation of action potential duration (APD) alternans at slow heart rates of 400 ms. The goal of this study is to find other cellular changes which play a role to the APD alternans at slow heart hearts. We conducted a single-cell simulation by scaling each parameter once independently and several combinations of scaled ionic parameters in the human ventricular model. For evaluating alternans, the model was paced from 750 to 180-ms cycle length (CL), and APD alternans in all CL was analyzed. In conclusion, combining scaled ionic parameters was able to induce the occurrence of APD alternans at slow heart rates compared to scaling the parameter once independently.
